I just got laid off from my job–who provided my health insurance.

Do I now have to sign up for Obamacare or pay a penalty of some sort? Have I missed the window to apply for healthcare this year?

I live in Florida, if that makes a difference.

If you are laid off, or simply lose employer based coverage, you have a limited time to enroll in a Marketplace plan or COBRA if it's offered. Learn more about Special Enrollment due to losing employer-sponsored coverage.

This is true in all states. Some states also offer Medicaid as an option. Medicaid coverage is based on income in many states, in other states (like Florida) there are more narrow guidelines.
